ID2 was detected in immersion fixed A549 human lung carcinoma cell line using 10 µg/mL Human ID2 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF4660) for 3 hours at room temperature. Cells were stained with the NorthernLights™ 557-conjugated Anti-Goat IgG Secondary Antibody (red, upper panel; Catalog # NL001) and counterstained with DAPI (blue, lower panel). Detection of Human ID2 by Western Blot.

Western blot shows lysates of HeLa human cervical epithelial carcinoma cell line. PVDF membrane was probed with 1 µg/mL of Human ID2 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF4660) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF019). A specific band was detected for ID2 at approximately 15 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 8.

Background: ID2

ID2 (Inhibitor of DNA binding 2; also known as inhibitor of differentiation 2) is a 15 kDa member of the small ID HLH family of proteins. It is a nuclear protein that forms inactivating heterodimers with bHLH transcription factors. bHLH proteins activate lineage-specific genes and ID2. Human ID2 is 134 amino acids (aa) in length. It contains one HLH domain (aa 36-76) but no DNA-binding motif. Full-length human ID2 is 98% aa identical to both mouse and porcine ID2.
