SMARCA5/SNF2H in Human Spleen.
SMARCA5/SNF2H was detected in immersion fixed paraffin-embedded sections of human spleen using Goat Anti-Human SMARCA5/SNF2H Antigen Affinity-purified Polyclonal Antibody (Catalog # AF6825) at 10 µg/mL overnight at 4 °C. Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (Catalog # CTS013). Tissue was stained using the Anti-Goat HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS008) and counterstained with hematoxylin (blue). Background: SMARCA5/SNF2H
SMARCA5 (Swi/SNF-related matrix-associated actin-dependent regulator of chromatin, subfamily A, member 5; also SNF2H) is a 130-135 kDa member of the SNF2/RAD54 helicase family of molecules. It is a ubiquitously expressed, chromatin remodeling ATPase that increases the accessibility of nucleosomal DNA to DNA-binding proteins. It apparently does so by sliding histone octamers over DNA. SMARCA5 is a component of four chromatin-remodeling complexes, including CHRAC, RSF, ACF and WICH. Human SMARCA5 is 1052 amino acids (aa) in length. It contains an N-terminal helicase domain (aa 192-357), a helicase superfamily C-terminal domain (aa 474-605), and two SANT DNA-binding domains (aa 840-892 and 943-1007). Over aa 77-185, human SMARCA5 shares 99% aa identity with mouse SMARCA5.
