SorLA in Human Brain.

SorLA was detected in immersion fixed paraffin-embedded sections of human brain (cortex) using Mouse Anti-Human SorLA Monoclonal Antibody (Catalog # MAB5699) at 25 µg/mL overnight at 4 °C. Before incubation with the primary antibody tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (Catalog # CTS013). Tissue was stained using the Anti-Mouse HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS002) and counter-stained with hematoxylin (blue). SorLA was detected in immersion fixed paraffin-embedded sections of human brain (cortex) using Mouse Anti-Human SorLA Monoclonal Antibody (Catalog # MAB5699) at 15 µg/mL overnight at 4 °C. Tissue was stained using the Anti-Mouse HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS002) and counterstained with hematoxylin (blue). Lower panel shows a lack of labeling if primary antibodies are omitted and tissue is stained only with secondary antibody followed by incubation with detection reagents. Background: SorLA

SorLA (sortilin-related receptor, LDLR class A repeats-containing), also called LR11 or SORL1, is a 250 kDa type I transmembrane glycoprotein of the Sortilin family of Vps10p-domain receptors. SorLA is found in both intracellular and surface membranes in the central nervous system, and mediates trafficking of proteins such as the amyloid precursor protein (APP). The 2214 amino acid (aa) human SorLA includes a 28 aa signal sequence, a 53 aa furin-cleaved propeptide, and a 2056 aa extracellular domain. The region used as an immunogen, which includes BNR, LDLR class A and B, and EGF-like domains, but excludes fibronectin-like domains, shares 95% aa identity with mouse and rat SorLA.
