Background: Stabilin-1
Stabilin-1, also known as FEEL-1 (fasciclin, EGF-like, laminin type EGF-like, and link domain containing scavenger receptor 1) and CLEVER-1 (common lymphatic endothelial and vascular endothelial receptor 1), was originally identified as the MS-1 antigen. It is a type I transmembrane multi-domain protein with a large extracellular domain that contains 7 fasciclin, multiple EGF-like and laminin type EGF-like, and a link domain. The short cytoplasmic tail interacts with CGAs which are cargo adaptors that link cargo into the clathrin-coated pits. Stabilin-1 is expressed by tissue macrophages and sinusoidal endothelial cells, which are scavenging cells. Stabilin-1 is involved in intracellular trafficking pathways and binds multiple ligands including acetylated LDL, SPARC, AGE, and SI-CLP. By alternative splicing, a soluble Stabilin-1 containing a 58 residue substitution between amino acid residues 746 and 803, followed by truncation, has been identified. Within the region used as immunogen, human Stabilin-1 shares 83% and 84% amino acid sequence homology with mouse and canine Stabilin-1, respectively.
