Background: IMP Dehydrogenase 2/IMPDH2

IMPDH, inosine monophosphate dehydrogenase, has been demonstrated to mediate negative regulation of Toll-like receptor 2 activity via the NF-kB signaling pathway (1). IMPDH upregulates phosphatase activity of SHP-1 which then goes on to inactivate PI3-k. The central function of IMPDH in the regulation of guanosine nucleotide pathways- conversion of IMP- inosine monophosphate to XMP-xanthosine monophosphate is a critical controlling step in de novo synthesis of guanosine nucleotides in T and B lymphocytes affecting proliferation (2). Because of this, IMPDH specific detection and control has made it a target for immunosuppressive, anti-cancer, anti-parasitic, or anti-viral therapy (3). Monitoring IMPDH or IMPDH2 activity as a biomarker for mycophenolic acid-induced immunosuppression in renal transplants is one such use of IMPDH or IMPDH2 specific probes (4).
