Recognizes a protein of 42-46kDa, identified MAGEA1 (MAGE-1). This monoclonal antibody does not cross-react with MAGE-2, -3, -4, -6 -9, -10, -or -12 protein. Human malignant neoplasms carry rejection antigens that are recognized by the patients' autologous, tumor directed and specific, cytolytic, CD8+ T lymphocyte clones (CTL). The MAGE family of genes codes an important group of antigens. It was identified that melanomas and primary glial brain tumors express common melanoma associated antigens (MAAs). Because MAGEA1 is expressed on a significant proportion of human neoplasms of various histological types (melanoma, brain tumors of glial origin, neuroblastoma, non-small cell lung cancer, breast, gastric, colorectal, ovarian, renal cell carcinomas) and not on normal tissues, the encoded antigen may serve as a marker of early detection and target for cancer immunotherapy.

Application Notes
Immunohistochemistry (Formalin-fixed): 1-2ug/ml for 30 minutes at RT. Staining of formalin-fixed tissues requires heating tissue sections in 10mM Tris with 1mM EDTA, pH 9.0, for 45 min at 95C followed by cooling at RT for 20 minutes.
Optimal dilution for a specific application should be determined.
Use in Sandwich -ELISA reported in scientific literature (PMID:31640756).
Please Note: Optimal dilutions of this antibody should be experimentally determined.

Background: MAGEA1

MAGE-1 is a cancer testis antigen (CTA) expressing in a variety of human cancers (Pereira CM et al., 2012). It is a human melanoma antigen recognized by cytolytic T lymphocytes. MAGE-1 induces a tumor specific immune response so it is thought to be a potential therapeutic target for cancer immunotherapy (Ma ZY et al., 2002). The gene for MAGE-1 belongs to a family of 12 genes known as MAGE-A cluster located on the X chromosome in region q28 (Laduron S et al., 2004). This relies primarily on DNA methylation for repression in somatic tissues but in many types of tumors, the promoter of these genes becomes demethylated and transcription becomes activated (Loriot A et al., 2006). Monoclonal antibody aganist a recombinant feline melanoma antigen 1 (fMAGE-1) has been generated and is effective against feline lymphoma cell lines and tumor tissues (Ma ZY et al., 2002).
