Background: NPM1

Nucleophosmin (NPM; also known as B23, numatrin or NO38) is a highly conserved phosphoprotein that belongs to nucleoplasmin/nucleophosmin family of nuclear chaperones. Expressed ubiquitously with nucleolar localization, NPM shuttles between nucleoli and cytoplasm during cell cycle progression. It binds ribosome to drive ribosome nuclear export and has been associated with nucleolar ribonucleoprotein structures. Chaperonin are the proteins that provide favourable conditions for the correct folding of other proteins for preventing aggregation; and NPM has the ability to acts as chaperonin for the core histones H3, H2B and H4 proteins. NPM stimulates APEX1 endonuclease activity on A/P double-stranded DNA while inhibit the same on A/P single-stranded RNA, and regulate APEX1 endonuclease activity within nucleoli devoted to repair AP on rDNA as well as the removal of oxidized rRNA molecules. BRCA2 helps NPM in regulating centrosome duplication, whereas, PLK2 mediated phosphorylation controls the centriole duplication. NPM is implicated in diverse cellular processes including cell proliferation, and regulation of tumor suppressors such as p53/TP53 and ARF.
