Application Notes
The J116 antibody has been tested by flow cytometric analysis of human PD-1 transfected cells. This can be used at less than or equal to 1 ug per test. A test is defined as the amount (ug) of antibody that will stain a cell sample in a final volume of 100 uL. Cell number should be determined empirically but can range from 105 to 108 cells/test. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.

Background: PD-1

PD1 is an inhibitory cell membrane surface receptor from the immunoglobulin superfamily. Involved in the regulation of T cell function during immunity and tolerance, PD1 inhibits T cell effector functions upon ligand binding. The protein is expressed in pro-B-cells and is thought to play a role in their differentiation as well. Studies have also suggested that this gene product may also contribute to the prevention of autoimmune diseases.
