Description

For extended storage aliquot contents and freeze at -20C or below. Avoid cycles of freezing and thawing. Centrifuge product if not completely clear after standing at room

This product was prepared from monospecific antiserum by immunoaffinity chromatography using phosphorylated threonine peptide coupled to agarose followed by conjugation to horseradish peroxidase and subsequent purification. Assay by immunoelectrophoresis resulted in a single precipitin arc against anti-Rabbit IgG and anti-Peroxidase

Application Notes
This product is suitable for use in ELISA and western blotting assays. The antibody reacts specifically with phosphothreonine and shows minimal reactivity by ELISA and western blotting with proteins containing phosphotyrosine or phosphoserine residues. The antibody reacts with free phosphothreonine, phosphothreonine conjugated to carriers such as thyroglobulin or BSA, and detects the presence of phosphothreonine proteins in both unstimulated and stimulated cell lysates. Although not tested, this antibody is likely functional in RIA, flow cytometry and immunohistochemistry.

Background: Phosphothreonine

Changes in the serine/threonine phosphorylation state of a protein in response to various external stimuli can have profound effects on cellular signal transduction, apoptosis and carcinogenesis. The reagents, including phosphorylated protein/peptides, antibodies against the phosphospecific amino acid, are important tools to explore the activation of serine, threonine or tyrosine containing proteins. An aberrant protein phosphorylation is a hallmark of human disease, and the enzymes, particularly protein kinases, which control protein phosphorylation are recognized as a major new drug target family.
