Product Specifications

Immunogen

Recombinant human PIP protein fragment (around aa 41-146) (exact sequence is proprietary) (Uniprot: P12273)

Localization

Cytoplasmic (Golgi bodies) and Secreted

Specificity

It recognizes a protein of 15kDa, identified as Gross cystic disease fluid protein 15 (GCDFP-15). It is a major protein component of benign breast gross cysts. It is a known marker of breast cancer, as it is found in approximately 50% of all breast cancer specimens. GCDFP-15, also known as PIP, for prolactin inducible protein, is a prolactin and androgen controlled protein. This antibody is useful in the identification of metastatic breast carcinoma, or fluid analysis.

Protein Array: PIP Antibody (PIP/1571) [NBP2-53226] - Analysis of Protein Array containing more than 19,000 full-length human proteins using PIP Antibody (PIP/1571). Z- and S- Score: The Z-score represents the strength of a signal that a monoclonal antibody (MAb) (in combination with a fluorescently-tagged anti-IgG secondary antibody) produces when binding to a particular protein on the HuProt(TM) array. Z-scores are described in units of standard deviations (SD's) above the mean value of all signals generated on that array. If targets on HuProt(TM) are arranged in descending order of the Z-score, the S-score is the difference (also in units of SD's) between the Z-score. S-score therefore represents the relative target specificity of a MAb to its intended target. A MAb is considered to specific to its intended target, if the MAb has an S-score of at least 2.5.

Applications for PIP Antibody (PIP/1571)

Application
Recommended Usage

Flow Cytometry

1-2 ug/million cells

Immunocytochemistry/ Immunofluorescence

1-2 ug/ml

Immunohistochemistry-Paraffin

1-2 ug/ml

Western Blot

1-2 ug/ml
Application Notes
Immunohistochemistry (Formalin-fixed): 1-2ug/ml for 30 minutes at RT. Staining of formalin-fixed tissues is enhanced by heating tissue sections in 10mM Tris buffer with 1mM EDTA, pH 9.0, for 45 min at 95C followed by cooling at RT for 20 minutes.
Optimal dilution for a specific application should be determined.

Background: PIP

The androgen and prolactin responsive prolactin-inducible protein (PIP), also known as Gross cystic disease fluid protein (GCDFP-15) is expressed in benign and malignant human breast tumors and in such normal exocrine organs as sweat, salivary and lacrimal glands. The hormonally responsive PIP gene is expressed in benign and malignant breast tumor tissues and in such normal exocrine organs as sweat, salivary, and lacrimal glands. The cells within the body that produce PIP/GCDFP 15 appear to be restricted primarily to those with apocrine function. Studies have found PIP to be a highly specific and sensitive marker for breast cancer.
