A portion of amino acids 186-199 (PLPRGHRAPEMEPN) of human PUMA a was used as the immunogen.

Reactivity Notes

The amino acid sequence used as immunogen is 78% homologous in rat.

Specificity

The amino acid sequence used as immunogen is 100% homologous in human (PUMA a and b). Human PUMA a is a 193 amino acid protein with a predicted molecular weight of approximately 22 kD. Human PUMA b is a 131 amino acid protein with a predicted molecular weight of approximately 15 kD.

Background: PUMA

A novel p53 inducible pro-apoptotic gene was identified recently and designated PUMA (for p53 upregulated modulator of apoptosis) in human and mouse. The PUMA gene encodes two BH3 domain-containing proteins termed PUMA-alpha and PUMA-beta. PUMA proteins bind Bcl-2, localize to the mitochondria, and induce cytochrome c release and apoptosis in response to p53. PUMA may be a direct mediator of p53-induced apoptosis.
