A synthetic peptide (AAPPLTLEGPVQSHHGTP) corresponding to amino acids 36-53 of human PUMA d (GenBank no. gb|AAK39544.1|AF354656_1 ) and amino acids 196-213 of human PUMA g (GenBank no ref|NP_001120712.1|) was used as the immunogen.

Specificity

The amino acid sequence used as immunogen has 100% identity in PUMAd and PUMAg, and 59% identity in PUMAa. For additional information on PUMA isoforms and their sequences, see Nakano et al. 2001.

Background: PUMA

A novel p53 inducible pro-apoptotic gene was identified and designated PUMA (for p53 upregulated modulator of apoptosis) in human and mouse. The PUMA gene encodes two BH3 domain-containing proteins termed PUMA-a and PUMA-b. PUMA proteins bind Bcl-2, localize to the mitochondria, and induce cytochrome c release and apoptosis in response to p53. PUMA may be a direct mediator of p53-induced apoptosis.
