Immunocytochemistry/Immunofluorescence: PYGL Antibody [NBP2-97708] - Immunofluorescence staining of PYGL in U251MG cells. Cells were fixed with 4% PFA, permeabilzed with 0.1% Triton X-100 in PBS, blocked with 10% serum, and incubated with rabbit anti-Human PYGL polyclonal antibody (dilution ratio 1:200) at 4C overnight. Then cells were stained with the Alexa Fluor(R)488-conjugated Goat Anti-rabbit IgG secondary antibody (green) and counterstained with DAPI (blue).Positive staining was localized to Cytoplasm and cell membrane.

Background: PYGL

Phosphorylase is an important allosteric enzyme in carbohydrate metabolism. Enzymes from different sources differ in their regulatory mechanisms and in their natural substrates. However, all known phosphorylases share catalytic and structural properties. Activity of phosphorylase is controlled both by allosteric means (through the noncovalent binding of metabolites) and by covalent modification. Thus AMP allosterically activates, whereas ATP, ADP, and glucose 6 phosphate allosterically inhibit, phosphorylase B.
