Immunohistochemistry-Paraffin: S-arrestin Antibody (PDS-1) [NB100-2385] - S-arrestin was detected in immersion fixed paraffin-embedded sections of human prostate cancer using Mouse Anti-Human S-arrestin (PDS-1) Monoclonal Antibody (Catalog # NB100-2385) at 1:300 for 1 hour at room temperature followed by incubation with the Anti-Mouse IgG VisUCyte™ HRP Polymer Antibody (Catalog # VC001). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to the cytoplasm.

Background: S-arrestin

Arrestins family contains four types of soluble cytoplasmic proteins that act as negative regulators of GPCR signaling, wherein, cognate ligand binding stimulates GPCR phosphorylation, followed by arrestin binding to phospho-GPCR leading to receptor internalization and ultimately, the desensitization of GPCR signaling. Among four different types of mammalian arrestins, the visual proteins namely arrestin 1 (S-arrestin, S-antigen or SAG) and arrestin 4 (X-arrestin) are localized to retinal rods and cones, respectively, whereas the non-visual proteins arrestin 2 (beta-arrestin 1) and arrestin 3 (beta-arrestin 2) are ubiquitously expressed. S-arrestin is rod specific and constitutes major proteins of retinal rod outer segments and associate with photoactivated/phosphorylated rhodopsin, thereby preventing transducin-mediated phosphodiesterase activation. Besides retinal rod photoreceptor cell outer segment, S-arrestin is also expressed in the pineal gland where its function is largely unknown. Arrestin knockout mice develop photoreceptor cell degeneration because of continuous phototransduction upregulation, as a consequence of defective rhodopsin/opsin shut-off mechanism, and in human subjects, S-arrestin mutations have been linked to Oguchi disease and retinitis pigmentosa.
