Immunohistochemistry: SARS-CoV-2 nsp7 Antibody [NBP3-07048] - Immunohistochemical analysis of paraffin-embedded COVID-19 patient lung tissue using anti- SARS-CoV-2 (COVID-19) NSP7 anbody (0.5 ug/mL). Tissue was fixed with formaldehyde and blocked with 10% serum for 1 h at RT; antigen retrieval was by heat mediation with a citrate buffer (pH6). Samples were incubated with primary antibody overnight at 4 degrees celsius. A goat anti-rabbit IgG H&L (HRP) at 1/250 was used as secondary. Counter stained with Hematoxylin. Strong signal of SARS-COV-2 NSP7 protein was observed in macrophage of COVID-19 patient lung, but not in non-COVID-19 patient lung.

Background: SARS-CoV-2 nsp7

SARS-CoV-2 Nonstructural Protein 7 (NSP7) is one of the sixteen nonstructural proteins of severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2), the causative agent of COVID-19 (1). SARS-CoV-2 NSP7 is 83 amino acids (aa) with a theoretical molecular weight of 9.2 kDa (1,2). The amino acid sequence alignment of SARS-CoV and SARS-CoV-2's NSP7 has 98.8% sequence identity and 100% sequence similarity (1). Functionally, SARS-Cov-2 NSP7 forms a complex with NSP8 and NSP12 to generate NSP8's RNA polymerase activity (1-3). In SARS-CoV, NSP7 and NSP8 are required for viral survival and form a channel-like, cylinder supercomplex (3). The NSP7-NSP8-NSP12 complex is considered the minimal core complex for RNA synthesis (4). While structurally similar to SARS-CoV, the components of the NSP7-NSP8-NSP12 complex of SARS-CoV-2 have reduced polymerase activity and thermostability (4).
