Background: SOCS-5

The suppressor of cytokine signaling (SOCS) family of proteins (SOCS1-SOCS7 and CIS) inhibit cytokine production by a classical negative feedback mechanism. These proteins are upregulated in response to cytokine stimulation followed by inhibition of cytokine-inducing signal pathway. Role of each member of the family has been determined by gene knockout experiments (1). SOCS5 protein is preferentially expressed in committed T-helper (Th1) cells. The action of SOCS5 is probably mediated by its interaction with the cytoplasmic domain of IL-4 receptor. During Th1 differentiation, there is a reduction in the association of Janus kinase 1 with the IL-4 receptor (IL-4R), which is correlated with the expression of SOCS5 pressor of cytokine signaling-5 (SOCS5). It has been also shown that in the overexpression of SOCS5 in transgenic mice leads to impaired Th2 development, suggesting that SOCS5 may plan an important role in the Th1 differentiation by regulating a balance between Th1 and Th2 (2).
