Immunoprecipitation:Sphingosine Kinase 1/SPHK1 AntibodyNBP3-29344] -Detection of human SPHK1 by western blot of immunoprecipitates. Samples: Whole cell lysate (1 mg for IP, 20% of IP loaded) from HeLa cells. Antibodies: Affinity purified rabbit anti-SPHK1 antibody NBP3-29344 used for IP at 10 µg/mg lysate. SPHK1 was also immunoprecipitated by rabbit anti-SPHK1., which recognizes a downstream epitope. For blotting immunoprecipitated SPHK1, A302-177A was used at 1 µg/ml. Detection: Chemiluminescence with an exposure time of 30 seconds. Note: The more slowly migrating bands are believed to represent additional forms of SPHK1 owing to recognition by multiple antibodies each of which was directed toward distinctly different epitopes on SPHK1.
