Background: VAV3

The Vav family are Rho/Rac guanosine nucleotide exchange factors (GEFs), consisting of three members in mammalian cells (Vav, Vav2, Vav3) and one in nematodes (CelVav) (1). Vav3 is expressed in most hematopoietic cells and non-hematopoietic tissues except ovary and skeletal muscles. T-cell stimulated and tyrosine phosphorylated Vav3 acts as an exchange factor for GTP-binding proteins RhoA, RhoG, and Rac1 (2). Also, tyrosine phosphorylated Vav3 mediates RTK signalizing, specifically IR, IGFR, and EGFR-mediated signaling pathways. Vav3 has been linked to as a factor for cell morphology modulation and an inducer for cell transformation (3).
