Application Notes

HeLa whole cell lysate is provided as a Western blot positive control. The lysates are provided as a set of treated and untreated samples. Use 10 ul per lane for a standard mini-gel blot (1.0 mg/ml).

Formulation, Preparation, and Storage

Preparation Method

HeLa cells are grown under standard tissue culture conditions until semi-confluent (70-80%). The cells are then treated with or without 10 mM DTT for 1 hour, washed in PBS and directly lysed into 1X Laemmli sample buffer with BME. The lysate is sonicated and tested by Western blot.

Formulation

The protein lysate is prepared in 1x Laemmli sample buffer with BME. Boiling before loading is not necessary

Background: HeLa

HeLa cells are the oldest and most commonly used human cell line in research. HeLa was derived from cervical cancer cells taken on February 9, 1951 from Henrietta Lacks. They were immortalized by George Otto Gey at Johns Hopkins University. HeLa was used by Jonas Salk to test the polio vaccine in the 1950s and have been used extensively in research since then, having been used in over 60,000 publications to date. In March of 2013, the HeLa genome was sequenced.
