Background: EGF-L6

Epidermal Growth Factor-like Domain, Multiple 6 (EGF-L6), also known as MAM- and EGF-containing (MAEG) is a member of the EGF repeat superfamily of proteins. This secreted protein contains five EGF-like repeats, an RGD motif that binds to Integrin alpha8 beta1, and a C-terminal MAM domain (1, 2). Within shared regions, human EGF-L6 shares 75% and 78% amino acid sequence identity with the mouse and rat orthologs, respectively. EGF-L6 is expressed in a multitude of fetal tissues during early development such as lung, heart, liver, spleen, cochlea, and placenta (3, 4). It is also expressed in the dermatome and dermatome-derived tissues including trunk dermis, hair follicles, and cranio-facial mesenchyme (5). EGF-L6 is expressed by osteoblastic-like cells and has been shown to act in a paracrine manner to promote migration of vascular endothelial cells, thus regulating angiogenesis in the local environment of the bone (4, 6). Additionally, EGF-L6 is expressed by epidermal stem cells in the hair follicle bulge and is believed to play a role in regulating the dermal vasculature (7). EGF-L6 is also produced by mature adipocytes and is thought to be involved in the process of adipose tissue expansion and the development of obesity (8). EGF-L6 expression is increased in fibroblastic meningioma and ovarian cancer and has been proposed as a biomarker for the latter (9, 10).
