Background: ISLR-2

ISLR2 (Immunoglobulin Superfamily containing Leucine-rich Repeat protein 2; also known as LINX/Leucine-rich repeat domain and Immunoglobulin domain containing axoN eXtension protein and Mbu-3 [in mouse]) is a member of the LIG superfamily of molecules (1, 2). Members of this family are typically integral membrane proteins that are characterized by the presence of multiple LRRs (leucine-rich repeats) N-terminal to one Ig domain.  ISLR2 should not be confused with an identically‑named, widely-expressed transcript isoform containing the ISLR1 protein (3). ISLR2 is principally associated with the embryonic nervous system.  In particular, it appears to coexist with TRKs on both sensory and motor postmitotic neurons (2, 4, 5). On these spinal neuron cell types, ISLR2 (plus other LIG family members) is suggested to fine-tune TrkA, TrkC and Ret downstream signaling in a temporally-restricted manner. This is posited to impact axonal outgrowth, branching and fasciculation (5). Notably, a lack of ISLR2 does not abrogate TrkA signaling. This suggests a modulating role for the molecule, and is consistent with the observation that ISLR2 and Trk interact through their extracellular domains (4, 5). Mature human ISLR2 is a 100-120 kDa, 727 amino acid (aa) type I transmembrane protein. It contains a 571 aa extracellular region (aa 19-589) plus a 135 aa cytoplasmic domain (2, 6). The extracellular region possesses seven LRR repeats (aa 19‑232) coupled to one Ig-like domain (aa 233-371).  Human and mouse extracellular domains share 89% aa sequence  identity (6).
