Background: SEZ6L

Seizure 6-like Protein (SEZ6L), also known as Brain-specific Receptor-like Protein B (BSRP-B), is a member of the SEZ6 family of proteins with a molecular weight of approximately 200 kDa (1). Mouse SEZ6L is a type I transmembrane glycoprotein composed of an extracellular domain (ECD) that contains three CUB domains and five short consensus repeats (SCRs) (1). Alternative splicing generates an isoform that contains a phenylalanine substitution for amino acids (aa) 807-872 of the fifth SCR domain (1). Mouse SEZ6L shares 80% and 97% aa sequence identity with the human and rat orthologs, respectively. It is expressed in the plasma membrane and endoplasmic reticulum of neurons and lung bronchiolar epithelium and likely regulates PKC alpha activation (1, 2). In mouse pancreatic beta cells, SEZ6L has been shown to be a substrate for BACE-2, which cleaves its ECD at a site 16 aa upstream from its transmembrane domain (3). Additionally, human SEZ6L has been shown to be involved in the maintenance of cellular cholesterol homeostasis (4). BSRP-C also known as SEZ6, has been reported to inhibit neurite outgrowth in mice (5). R&D Systems in-house testing shows that mouse SEZ6L, also exhibits this function. Genetic variations in human Sez6l have been associated with bipolar disorder I and an increased risk for lung cancer (2, 6, 7).
