Background: SESN2

Sestrin 2(SESN2), located at human chromosome 1p35.3, belongs to the sestrin family of proteins. These proteins are cystein sulfinyl reductases and they modulate peroxide signaling and antioxidant defense (1). This family of proteins is only present in multicellular organisms ranging from nematodes to mammals. These proteins selectively reduce or repair hyperoxidized forms of typical 2-Cys peroxiredoxins within eukaryotes (2). Peroxiredoxins are the enzymes that metabolize peroxides. Expression of these proteins is regulated by p53, a tumor suppressor protein (3). Recently it was reported that sestrin 2 is not a sulfinic peroxiredoxin reductase (4). Sestrin 2 is implicated in the regulation of cell growth and survival, in cellular responses to different stress conditions, and also acts as a positive regulator of autophagy (5). Depletion of sestrin 2 in human cancer cells reduced the level of autophagy when autophagy was induced in these cells (5). Similar to sestrin 1, sestrin 2 negatively regulates the activity of mammalian target of rapamycin (mTOR), a positive regulator of cell growth (6). Sestrin 2 may be a promising target in the treatment of chronic obstructive pulmonary disease (COPD). It was reported that mutational inactivation of sestrin 2 induced TGF-Beta signaling and partially reduced the emphysema phenotype in mice model of COPD (7). Sestrin 2 might also be a key target in the treatment of cancer. It was shown to activate AMPK and repress mTORC1 signaling. AMPK is an essential mediator of LKB1 tumor suppressor and it is down-regulated in cancer cells (8). Sestrin 2 is approximately a 57.6kDa protein based on the amino acid predicted MW of 57kDa (480 amino acids).
