Charge Characterization of Adeno-Associated Virus (AAV) Capsid Proteins Using icIEF

Scientific Meeting PostersPost-translational modifications to individual capsid proteins of AAVs have been shown to reduce transduction efficiency, therefore assessing the stability of these capsid proteins is critical for therapeutic development. This poster presents a rapid charge characterization method for AAV capsid proteins based on icIEF using the Maurice (native fluorescence detection) and Peggy Sue (chemiluminescence detection) systems. The method demonstrates:
- The analysis of eight different AAV serotypes, capturing amino acid differences as little as 1% and serving as an identity assay
- The stability-indicating property of icIEF on these systems, providing a rapid way to detect subtle changes in capsid proteins in pure or complex systems
