Neurofilament Light (NF-L) in Cerebrospinal Fluid, Serum, and Plasma

Neurofilament light chain has been implicated as an important biomarker for a number of neurological disorders. The Simple Plex neurofilament light assay can quantify this biomarker in serum and plasma samples to provide highly robust and reproducible data.

The Simple Plex Assay Correlates with the Uman Diagnostics ELISA

 

Cerebrospinal fluid (CSF) NF-L levels measured using the Simple Plex Assay correlate with those quantified by the Uman Diagnostics ELISA. Samples were prepared according to the recommendations of the respective kit manufacturer. Levels of NF-L were measured within the linear range of each assay. The dilution factors were used to back-calculate the original concentration in the samples. These values were then plotted against each other, and a linear curve-fitting algorithm was applied.

The Simple Plex Assay Correlates with the Simoa Bead-Based Assay

 

Plasma/serum NF-L levels measured using the Simple Plex Assay correlate highly with the Simoa assay. Samples were prepared according to the manufacturer's instructions. NF-L levels were measured within the linear range of each assay. The dilution factors were used to back-calculate the initial concentration in the samples. These values were then plotted against each other, and a linear curve-fitting algorithm was applied.

The same plasma and serum samples, obtained from healthy donors, were prepared according to the Simple Plex NF-L Assay instructions and measured at three different sites. The mean sample coefficient of variation (CV) was less than 10%.

NF-L is Readily Detectable in CSF, Plasma and Serum

 

The ability to detect neurofilament light in the blood is important to allow minimally-invasive diagnosis and monitoring of disease progression. NF-L is readily detectable in cerebrospinal fluid (CSF), serum, EDTA plasma, and Heparin plasma. Serum and CSF samples are 100% detectable. Plasma samples are 90% detectable.

The Simple Plex Assay Detects Elevated NF-L in Serum From ALS Patients

 

ALS patient samples were used as a model system to quantify NF-L levels since previous reports have established elevated NF-L levels in ALS serum. Healthy samples were analyzed on a 72x1 Simple Plex NF-L Cartridge and increased NF-L levels were observed in the ALS samples. Bars represent the median NF-L level ± the interquartile range. The statistical difference was evaluated using the nonparametric Mann-Whitney test with 19 to 20 samples per group.
