Immunohistochemistry: ABCG2/CD338 Antibody (JF0994) [NBP3-31988] - Immunohistochemical analysis of paraffin-embedded mouse placenta tissue using anti-ABCG2/CD338 antibody. The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 8.0-8.4) for 20 minutes.The tissues were blocked in 5% BSA for 30 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (NBP3-31988, 1/50) for 30 minutes at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Immunocytochemistry/ Immunofluorescence: ABCG2/CD338 Antibody (JF0994) [NBP3-31988] - ICC staining of ABCG2/CD338 in Hela cells (red). Formalin fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 10 minutes at room temperature and blocked with 1% Blocker BSA for 15 minutes at room temperature. Cells were probed with the primary antibody (NBP3-31988, 1/50) for 1 hour at room temperature, washed with PBS. Alexa Fluor®594 Goat anti-Rabbit IgG was used as the secondary antibody at 1/1,000 dilution. The nuclear counter stain is DAPI (blue).

Flow Cytometry: ABCG2/CD338 Antibody (JF0994) [NBP3-31988] - Flow cytometric analysis of ABCG2/CD338 was done on 293T cells. The cells were fixed, permeabilized and stained with the primary antibody (NBP3-31988, 1/50) (red). After incubation of the primary antibody at room temperature for an hour, the cells were stained with a Alexa Fluor 488-conjugated Goat anti-Rabbit IgG Secondary antibody at 1/1000 dilution for 30 minutes.Unlabelled sample was used as a control (cells without incubation with primary antibody; black).

Background: ABCG2

Tumor cells may display a multidrug resistant phenotype by overexpression of ATP-binding cassette transporters such as multidrug resistance (MDRI) p-glycoprotein, multidrug resistance protein 1 (MRP1), and breast cancer resistance protein (BCRP). Tumor cells can be intrinsically resistant to drugs or they can acquire resistance to structurally and functionally unrelated drugs on drug exposure. This phenomenon is known as multidrug resistance (MDR). In human tumor cells, several transporter proteins can be involved in MDR. These proteins, MDR1 P-gp (ABCB1), MRP1 (ABCC1), MRP2 (ABCC2), MRP3 (ABCC3), and BCRP (ABCG2), belong to the ABC transporter family. They act as efflux pumps, which result in decreased intracellular concentrations of cytotoxic drugs. BCRP is a recently discovered half-transporter that probably acts as a homo- or heterodimer in transporting cytotoxic agents. The transporter molecule is capable of transporting several anticancer drugs but has thus far been found mainly in MX-resistant cell lines.
