Scientific Data Images for Alpha Fodrin Antibody (3D7)

Western Blot: Alpha Fodrin Antibody (3D7) [NBP1-92689]

Western Blot: Alpha Fodrin Antibody (3D7) [NBP1-92689]

Western Blot: Alpha Fodrin Antibody (3D7) [NBP1-92689] - Analysis of neural tissue and cell lysates using mouse mAb to alpha-II spectrin (NBP1-92689, green). [1] protein standard, [2] rat whole brain, [3] rat spinal cord, [4] mouse whole brain, [5] mouse spinal cord, [6] NIH-3T3, [7] HEK293, [8] HeLa, [9] SH-SY5Y, [10] C6 cells. A prominent band at about 250-260 kDa represents the intact alpha-II spectrin heavy chain in neural tissues or cell lines, but not in HeLa.
Immunocytochemistry/ Immunofluorescence: Alpha Fodrin Antibody (3D7) [NBP1-92689]

Immunocytochemistry/ Immunofluorescence: Alpha Fodrin Antibody (3D7) [NBP1-92689]

Immunocytochemistry/Immunofluorescence: Alpha Fodrin Antibody (3D7) [NBP1-92689] - Confocal image of mixed neuron-glial cultures stained with NBP1-92689 antibody to Alpha Fodrin (green) and counterstained for DNA (blue). The Alpha Fodrin antibody stains numerous axonal and dendritic profiles in these cultures, and this image shows an optical section through a group of neuronal cell bodies. NBP1-92689 clearly reveals the submembraneous cytoskeleton. Since Alpha Fodrin is specific for neurons in the CNS, the glial cells in this culture are not recognized by this antibody. This antibody also reveals the submembranous cytoskeleton of the axon.
Immunohistochemistry: Alpha Fodrin Antibody (3D7) [NBP1-92689]

Immunohistochemistry: Alpha Fodrin Antibody (3D7) [NBP1-92689]

Immunohistochemistry: Alpha Fodrin Antibody (3D7) [NBP1-92689] - Immunofluorescent image of rat cerebellum stained with NBP1-92689 antibody to alpha-II spectrin, dilution 1:2,000, in green, chicken polyclonal antibody to GFAP, dilution 1:5,000, in red. The spectrin antibody stains the submembraneous cytoskeleton on neurons and strongly reveals the cell bodies and dendrites of Purkinje cells, while the GFAP antibody stains the processes of Bergmann glia and astrocytes.

Background: Alpha Fodrin

Alpha Fodrin (SPTAN1; also called non-erythroid alpha spectrin) belongs to spectrin family of proteins which are widely distributed filamentous proteins implicated in generating a supporting/organizing scaffold for cell membranes, and SPTAN1 protein is involved in processes such as secretion, interactions with calmodulin in Ca2+ dependent manner and Ca2+ dependent cytoskeletal movements at the membranes. SPTAN1 interacts with several proteins including ACP1, CALM, EMD, TRPC4 etc. and forms a component of a complex that contains several other proteins such as ACTN4, CASK, IQGAP1, MAGI2, NPHS1 and SPTBN1. SPTAN1 cleavage accompanies apoptosis, induced by activation via CD3/TCR complex, Fas/CD95 ligation and other Fas-expressing cells or through cellular exposure to staurosporine, dexamethasone, synthetic ceramide etc. Major products of SPTAN1 cleavage are N-terminal/C-terminal fragments, and phosphorylation of Tyr-1176 has been shown to decrease its sensitivity to cleavage by calpain in vitro. Defects in SPTAN1 have been linked to EIEE5 (epileptic encephalopathy early infantile type 5).
