Western Blot: CCR8 Antibody (JE31-99) [NBP3-32117]

Western Blot: CCR8 Antibody (JE31-99) [NBP3-32117] - Western blot analysis of CCR8 on mouse thymus tissue lysates with Rabbit anti-CCR8 antibody (NBP3-32117) at 1/1,000 dilution.

Lysates/proteins at 30 ug/Lane.

Predicted band size: 41 kDa
Observed band size: 48 kDa

Exposure time: 3 minutes;

4-20% SDS-PAGE gel.

Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (NBP3-32117) at 1/1,000 dilution was used in 5% NFDM/TBST at room temperature for 2 hours. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1:100,000 dilution was used for 1 hour at room temperature.

Immunocytochemistry/ Immunofluorescence: CCR8 Antibody (JE31-99) [NBP3-32117] - Immunocytochemistry analysis of Raji cells labeling CCR8 with Rabbit anti-CCR8 antibody (NBP3-32117) at 1/50 dilution.

Cells were fixed in 4% paraformaldehyde for 10 minutes at 37 ℃, permeabilized with 0.05% Triton X-100 in PBS for 20 minutes, and then blocked with 2% negative goat serum for 30 minutes at room temperature. Cells were then incubated with Rabbit anti-CCR8 antibody (NBP3-32117) at 1/50 dilution in 2% negative goat serum overnight at 4 ℃. Goat Anti-Rabbit IgG H&L (iFluor™ 488) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. Nuclear DNA was labelled in blue with DAPI.

Background: CCR8

CKR8/CCR8, or C-C chemokine receptor type 8, is a G protein-coupled, seven-transmembrane domain receptor protein. Chemokines are key molecules in directing leukocyte migration toward sites of inflammation. The human C-C chemokine I-309 is a monocyte chemoattractant and inhibits apoptosis in thymic cell lines. It has been found that CKR8/CCR8 is a receptor for chemokines SCYA1/I-309, SCYA4/MIP-1-beta, and SCYA17/TARC (1-3).
