Recognizes a protein of 210-220kDa, which is identified as the complement receptor 1 (CR1)/CD35. This monoclonal antibody is specific for a site in CR1 that is distal from the C3b-binding site, so that it is unable to block CR1 activity. This monoclonal antibody is highly specific to CR1 and shows no cross-reaction with CR2. The primary function of CR1 is to serve as the cellular receptor for C3b and C4b, the most important components of the complement system leading to clearance of foreign macromolecules. The Knops blood group system is a system of antigens located on this protein. Follicular dendritic cells (FDC) are restricted to the B-cell regions of secondary lymphoid follicles. They are CD21+/CD35+/CD1a-. This monoclonal antibody labels follicular dendritic cells and follicular dendritic cell sarcoma.

Application Notes
Immunohistochemistry (Formalin-fixed): 1-2ug/ml for 30 minutes at RT. Staining of formalin-fixed tissues requires heating tissue sections in 10mM Tris with 1mM EDTA, pH 9.0, for 45 min at 95C followed by cooling at RT for 20 minutes.
Optimal dilution for a specific application should be determined.
Please Note: Optimal dilutions of this antibody should be experimentally determined.

Background: CD35

CD35 encodes a membrane glycoprotein found on peripheral blood cells, glomerular podocytes, and follicular dendritic cells. The protein is a receptor for complement components C3b and C4b and regulates the activity of the complement cascade. Variation in this protein is the basis of the Knops blood group system. The two most common alleles, F and S, differ by 8 exons and are thought to be the result of an unequal crossover event. A secreted form of the protein present in plasma has been described, but its full length nature has not been determined.
