Background: Ceramide Kinase

Ceramide kinase (CERK) belongs to diacylglycerol kinase family of lipid kinases and it catalyzes specifically the phosphorylation of ceramide to generate ceramide 1-phosphate (C1P). CERK acts primarily on natural as well as analog ceramides such as C6, C8, C16 ceramides, and C8-dihydroceramide, to a lesser extent on C2-ceramide and C6-dihydroceramide, but not on other lipids including sphingosines. Localized in the cytoplasm and membrane as peripheral membrane protein, CERK is the only enzyme known to produce C1P, a signaling molecule which shows positive effect on cell proliferation or survival and also regulates cytosolic phospholipase A2 (cPLA2) activity . CERK/C1P signaling is implicated in mast cell activation, eicosanoid signaling, apoptosis, cellular immune response/inflammation, emotional behavior, angiogenesis, phagolysosome formation etc.
