Immunocytochemistry/Immunofluorescence: CNPase Antibody [NBP3-05551] - Immunofluorescent analysis of a section of mouse striatum stained with CNPase Antibody at a dilution of 1:3,000 in green, and costained with rabbit pAb to NF-L at a dilution of 1:5,000, in red. The blue is Hoechst staining of nuclear DNA. Following transcardial perfusion of mouse with 4% paraformaldehyde, the brain was post fixed for 24 hours, cut to 45uM, and free-floating sections were stained with above antibodies. The CNPase antibody stains myelin sheath and the plasma membranes of oligodendrocytes, the myelin producing cells of the CNS. The NF-L antibody labels axons of neuronal cells enclosed by the myelin.

Background: CNPase

2,3-cyclic nucleotide-3-phosphodiesterase (CNP) is a membrane bound, microtubule associated protein that is among the most abundant myelin proteins of the CNS. It is thought that CNP may serve as a regulator of tubulin polymerization and of microtubule distribution (Bifulco et al., 2002). It was recently found that CNP may also function as a possible linker protein anchoring microtubules to the plasma membrane via a 13 residue C-terminal CNP fragment (Bifulco et al., 2002, Esposito et al., 2008).
