Western Blot: CRABP2 Antibody (JE65-66) [NBP3-32214] - Western blot analysis of CRABP2 on MCF-7 cell lysates with Rabbit anti-CRABP2 antibody (NBP3-32214) at 1/1,000 dilution.

15% SDS-PAGE gel.

Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (NBP3-32214) at 1/1,000 dilution was used in 5% NFDM/TBST at room temperature for 2 hours. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1:300,000 dilution was used for 1 hour at room temperature.

Immunohistochemistry: CRABP2 Antibody (JE65-66) [NBP3-32214] - Immunohistochemical analysis of paraffin-embedded human esophagus tissue with Rabbit anti-CRABP2 antibody (NBP3-32214) at 1/400 dilution.

The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 20 minutes. The tissues were blocked in 1% BSA for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (NBP3-32214) at 1/400 dilution for 1 hour at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Background: CRABP2

Cellular retinoic acid binding protein 2 (CRABP2) is a coactivator present in the retinoic acid (RA)-dependent nuclear complex. CRABP2 has been shown to mediate retinoic acid activity in skin to influence skin cell growth and differentiation. It has also been shown to mediate the growth suppressive effects of retinoic acid in breast cancer. CRABP2's role in cell differentiation may be linked to control of the cell-cycle since it has been identified as an interacting partner of cyclin D3 in CRABPII and RAR ternary complex. Alternate names for CRABP2 include cellular retinoic acid-binding protein II, CRABP-II, retinoic acid-binding protein II, and RBP6.
