This antibody was raised by immunizing BALB/c mice with a BSA-conjugated peptide comprising of the N-terminal part of CXCR1/IL-8RA (amino acids 1-30).

Specificity

SE2 antibody is specific for CXCR1/IL-8RA and does not corss-react with IL-8RB. CXCR1/IL-8RA is a receptor to interleukin-8, which is a powerful neutrophils chemotactic factor. Binding of IL-8 to the receptor causes activation of neutrophils.

Western Blot: CXCR1/IL-8RA Antibody (SE2) - Chimeric [NBP3-12003] - Human lymph node tissue lysates (35ug protein in RIPA buffer) were resolved on a SDS PAGE gel and blots were probed with the chimeric rabbit version of SE2 (NBP3-12003) at 0.1 ug/ml before detection using an anti-rabbit secondary antibody. A primary incubation of 1h was used and protein was detected by chemiluminescence.

Immunocytochemistry/Immunofluorescence: CXCR1/IL-8RA Antibody (SE2) - Chimeric [NBP3-12003] - Immunofluorescence analysis of paraformaldehyde fixed human peripheral blood leukocytes on Shi-fix(TM) coverslips stained with the chimeric rabbit IgG version of SE2 (NBP3-12003) at 10 ug/ml for 1h followed by Alexa Fluor(R) 488 secondary antibody (2 ug/ml), showing membrane staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom NBP3-12003, DAPI, merged channels and an isotype control. The isotype control was an unknown specificity antibody followed by staining with Alexa Fluor(R) 488 secondary antibody.

Background: CXCR1/IL-8RA

CXCR1, also known as IL-8 RA and CD181, is an approximately 60 kDa 7TM glycoprotein that functions as a receptor for the chemokine CXCL8/IL-8. It is expressed on neutrophils, monocytes, CD8 T cells, FoxP3+ CD4 Treg cells, mast cells, neuronal and glial cells, vascular endothelial cells, and melanoma. CXCR1 forms homodimers and heterodimers with CXCR2/IL-8 RB. It can be cleaved from neutrophils in the lungs of cystic fibrosis patients to release fragments that promote CXCL8 production from airway epithelial cells. CXCR1 mediates neutrophil activation and chemotaxis to sites of inflammation as well as angiogenesis and melanoma invasiveness.
