Immunogen

MJ7/18 was prepared by immunizing rats with inflamed mouse skin tissue. The antibody was selected immunohistologically for reactivity with endothelial cells.

Specificity

MJ7/18 binds specifically to mouse Endoglin/CD105. Endoglin/CD105 is a homodimer expressed by vascular endothelial cells. Endoglin/CD105 functions in adhesion and embryonic angiogenesis. Endoglin/CD105 is a major homodimeric glycoprotein of vascular endothelium involved in cell ahesion through integrin/RGD binding and the regulation of angiogenesis. Also acts as TGF-beta coreceptor and is involved in the TGF-beta/BMP signaling cascade. Endoglin/CD105 is also required for GDF2/BMP9 signaling through SMAD1 in endothelial cells.

Immunocytochemistry/Immunofluorescence: Endoglin/CD105 Antibody (MJ7/18) - Chimeric [NBP2-81085] - Immunofluorescence staining of fixed mouse splenocytes with Endoglin/CD105 Antibody (MJ7/18) [NBP2-81085]. Immunofluorescence analysis of paraformaldehyde fixed mouse splenocytes on Shi-fix coverslips, permeabilized with 0.15% Triton and stained with the chimeric rabbit IgG version of MJ7/18 [NBP2-81085] at 10 ug/ml for 1h followed by Alexa Fluor 488 secondary antibody (1 ug/ml), showing membrane staining in a small subset of cells. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom NBP2-81085, DAPI, merged channels and an isotype control. The isotype control was stained with an anti-Fluorescein antibody followed by Alexa Fluor 488 secondary antibody.

Application Notes
This chimeric rabbit antibody was made using the variable domain sequences of the original Rat IgG2a format, for improved compatibility with existing reagents, assays and techniques. MJ7/18 can be used for FC, WB, IP and IHC (Baluk P, et al. 2003. Am. J. Pathol. 163:1801).

Background: Endoglin/CD105

CD105 is a 90 kD homodimeric type I integral membrane glycoprotein, also known as endoglin. It is expressed on endothelial cells (especially on angiogenic endothelial cells) and up-regulated by hypoxia, activated monocytes, macrophages, bone marrow stromal cells, and some cytotrophoblasts. CD105 is a receptor for TGF- szlig1, TGF- szlig3 and modulates TGF- szlig signaling by interacting with TGF- szlig receptors I and/or II. CD105 also binds other growth factors such as actvin A, BMP-2 and -7. CD105 has been show to be a useful marker for identifying proliferating endothelium involved in tumor angiogenesis and can be used for tumor imaging, prognosis, as well as has therapeutic potential for some solid tumors and other angiogenic diseases.
