Background: ErbB4/Her4

Epidermal Growth Factor (EGF) affects cell growth through its interaction with a 170 kDa cell surface glycoprotein, known as the EGF receptor. Binding of EGF or TGF alpha to the receptor activates its associated kinase. Other members of the EGF receptor family include Neu (ErbB 2, HER2), ErbB 3 (HER3) and ErbB 4 (HER4). The cytoplasmic regions of EGFR and Neu are quite similar, but ErbB 3 and ErbB4 more closely resemble each other than EGFR or Neu. ErbB4 is the fourth member of class I receptor kinase family. ErbB4 is most predominantly expressed in several breast carcinoma cell lines, and in normal skeletal muscle, heart, pituitary, brain, and cerebellum. Breast tumor cell lines T47D, MDA MB 453, BT 474 and H3396 are found to have the highest levels of mRNA, and intermediate levels are seen in MCF7, MDAMB 330 and MDA MB 361. Expression of ErbB4 is low or absent in some breast tumor cell lines such as MDA MB 231, MDA MB 157, MDA MB 468, and SKBR 3. Down-regulation of ErbB 4 in estrogen receptor-positive cancer cells inhibits proliferation.
