Staining of murine macrophages Murine bone marrow-derived macrophages (BMDMs) were stained with anti-Fc gamma RII/RIII (CD32/CD16) antibody (as well as isotype control) and bound antibody detected using goat IgG anti-rat IgG (H&L-chain) polyclonal antibody directly conjugated to Alexa Fluor 647(AF647) commercially available from a competitor. Compared to the isotype control and secondary-only control, a Fc gamma RII/RIII (CD32/CD16)-positive population is apparent.

x vivo murine (C57BL6/J) monocytes and neutrophils (from zymosan-elicited peritoneal exudate, left and middle panel) as well as bone marrow-derived macrophages (BMDMs, right panel) were stained with a murine anti-human antibody (fluorescently labelled (AF647) human-specific anti-CD16) with or without pre-incubation with anti-mouse CD16/CD32 (Fc-receptors) antibody (Ab00123-8.1, clone 2.4G2). Pre-incubation of cells with anti-CD16/CD32 reduced non-specific binding of the fluorescently labelled antibody to all cell types analysed and shows that this anti-CD16/CD32 antibody acts as an Fc-blocking reagent.

This antibody is Cytof ready. This chimeric rabbit antibody was made using the variable domain sequences of the original Rat IgG2b format, for improved compatibility with existing reagents, assays and techniques. This antibody binds murine Fc Receptors CD16/CD32 and inhibits binding of antibodies to these. It can therefore be used as an Fc-blocking reagent, to reduce non-specific staining of cells by antibodies binding via their Fc-region. A similar effect can be achieved using the Fc Silent range of antibodies that prevent these interactions at the level of the Fc receptor binding site, which has been mutated to lack affinity for the Fc receptors.

Background: Fc gamma RII/RIII (CD32/CD16)

Receptors that recognize the Fc portion of IgG are divided into three groups designated Fc gamma RI, RII, and RIII, also known respectively as CD64, CD32, and CD16. In humans each group of receptors is encoded by 2 - 3 closely related genes designated A, B, and C. Fc gamma RI binds IgG with high affinity and functions during early immune responses. Fc gamma RII and RIII are low affinity receptors that recognize IgG as aggregates surrounding multivalent antigens during late immune responses. Different Fc receptors can function as activators, as inhibitors, or as decoy receptors.
