Specificity

Follistatin is a single-chain glycosylated protein that inhibits follicle stimulating hormone (FSH) release. Alternative splicing of Follistatin mRNA yields two isoforms, FS315 and FS288. FS288 is the main cell-surface form and binds to surface heparin sulphate proteoglycans. FST/4281 is a clone raised against recombinant Fst 288, and is used in combination, commonly as the capture, with antibody FST/4282 in a two site ELISA for the detection of Follistatin.

Application Notes
ELISA: Use in combination with clone FST/4282 for two-site ELISA.
Immunohistochemistry (Formalin-fixed): 1-2ug/ml for 30 minutes at RT. Staining of formalin-fixed tissues requires heating tissue sections in 10mM Tris with 1mM EDTA, pH 9.0, for 45 min at 95C followed by cooling at RT for 20 minutes.
Optimal dilution for a specific application should be determined.

Background: Follistatin

FST, also known as Follistatin, has a 344 amino acid long isoform that is 38 kDa and a short 317 amino acid that is 35 kDa, functions as an activin antagonist binding directly to it and inhibits the biosynthesis and secretion of pituitary follicle stimulating hormone (FSH). Studies of this protein are being performed in relation to polycystic ovary syndrome, pituitary adenoma, insulin resistance, Down syndrome, plasmacytoma, lymphocytic leukemia, pituitary tumor infertility, endometrial carcinoma, teratocarcinoma, azoospermia, meningitis, hepatocellular carcinoma, ovarian carcinoma, liver disease, endometriosis, prostate carcinoma, prostatitis, ovarian cancer, and adenoma. This protein has been also shown to have interactions with ANG, DIP2A, BMP4, FN1, and TXN in signal transduction activin A signaling regulation and TGF-beta signaling pathways.
