Background: Galectin-3

Galectin-3 (LGALS3 or Mac-2 antigen), is a 26 kD beta-galactoside-binding protein from galectin family and it is ubiquitously expressed with localization dependent on cell type as well as proliferative status. Galectin-3 is expressed in many immunocompetent and inflammatory cells, including macrophages, dendritic cells, eosinophils, mast cells, uterine NK cells and activated T and B cells, and it is localized in nucleus, cytoplasm or on cell surface as well as in extracellular compartments. Extracellularly, galectin-3 acts as an adhesion molecule whereas intracellular galectin-3 implicates in processes that are pivotal for basic cellular functions, such as pre-mRNA splicing regulation of cell growth, cell cycle progression and apoptosis. Galectin-3 binds and interacts with numerous ligands in intra-/extracellular environment such as DMBT1, ITGA3, ITGB1, CSPG4, LGALS3BP, LYPD3, CYHR1, UACA etc. for modulating cellular signals responsible fpr its biological functions such as phagocytosis, cell growth/proliferation, adhesion and apoptosis in normal as well as cancer cells. Circulating galectin-3 concentrations increases more than 30-fold in bloodstream of patients with various cancers and patients with metastatic disease have higher concentrations of circulating galectin-3 than those with only localized tumors.
