Detection of Human APAF-1 by Western Blot.
Western blot shows lysates of Jurkat human acute T cell leukemia cell line. Gels were loaded with 2 x 105cells (lane 1) and 7 x 104cells (lane 2). PVDF membrane was probed with 1 µg/mL Mouse Anti-Human APAF-1 Monoclonal Antibody (Catalog # MAB868) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF007). A specific band for APAF-1 was detected at approximately 135 kDa (as indicated). Background: APAF-1
Apoptosis protease-activating factor 1 (APAF‑1) is a ubiquitous 135 kDa protein component of the cytoplasmic apoptosome complex. APAF‑1 plays an important role in the mitochondrial apoptotic pathway by triggering the Cytochrome c-dependent activation of Caspase-9. APAF‑1 participates in tissue morphogenesis during development, and loss of APAF‑1 expression is associated with tumor progression in a variety of cancers. Some alternate splice forms of APAF‑1 are deficient in their ability to promote apoptosis.
