APH1A was detected in immersion fixed paraffin-embedded sections of human Alzheimer's brain using Mouse Anti-Human APH1A Monoclonal Antibody (Catalog # MAB6908) at 15 µg/mL overnight at 4 °C. Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (Catalog # CTS013). Tissue was stained using the Anti-Mouse HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS002) and counterstained with hematoxylin (blue). Specific staining was localized to neurons. Background: APH1A

Anterior pharynx-defective 1 (APH1A) is a widely expressed 36 kDa 7-transmembrane protein found in the endoplasmic reticulum and Golgi. APH1A is a component of the gamma secretase proteolytic complex which cleaves multiple substrates within their transmembrane domains. This activity liberates the intracellular domains of ErbB4, Neuregulin-1, and Notch receptors in addition to the A-beta peptide from the amyloid precursor protein (APP). Alternate splicing generates a short isoform of human APH1A that lacks 18 amino acids at the C-terminus. Within aa 58-69, human APH1A shares 100% aa sequence identity with mouse and rat APH1A.
