Background: Complement Factor H-related 4/CFHR4
FHR4 (Complement factor H-related protein 4), also known as CFHR4, is an 84-86 kDa glycoprotein member of group 2 in the factor H-related protein family of molecules. It is synthesized by hepatocytes and circulates in plasma as a monomer. FHR4 binds to lipoproteins (LDL and VLDL), C3b and C3d, and to native (pentameric) C-reactive protein (pCRP). In addition to full-length 86 kDa CFHR4 (called FHR4A), there are two alternative splice forms. One simply shows a deletion of Glu20 relative to the full-length form. A second called FHR4B is 42-45 kDa in size, and shows a deletion of amino acids (aa) 60-306 within the mature CFHR4 molecule. This short isoform is the molecule used as the immunogen for clone 640212. This 312 aa short form contains five Sushi/SCR domains, the first of which (aa 23-85) is associated with pCRP binding. Both major isoforms appear to show the same activity. That is, they serve as a platform for C3 convertase assembly, and bind pCRP which leads to classical complement activation. Over aa 20-331, human CFHRB shares 47% aa identity with a related molecule found in rat.
