Human NK Cells either (A) stimulated in a 1:1 ratio with K562 human chronic myelogenous leukemia cell line and 3 µM monensin for 5 hours or (B) ustimulated were stained with Mouse Anti-Human LAMP-1/CD107a Alexa Fluor® 488-conjugated Monoclonal Antibody (Catalog # IC4800G) and Mouse Anti-Human NCAM-1/CD56 PE-conjugated Monoclonal Antibody (FAB2408P). Human NK Cells were expanded from PBMCs following the protocol on ExCellerate Human NK Cell Expansion Media, Xeno Free (CCM032) plus Recombinant Human IL-2 (27 ng/mL), Recombinant Human IL-12 (10 ng/mL), Recombinant Human IL-18 (10 ng/mL), and Recombinant Human IL-21 (10 ng/mL) for 14 days, then stained with (A) Mouse Anti-Human LAMP-1/CD107a Alexa Fluor® 488-conjugated Monoclonal Antibody (Catalog # IC4800G) or (B) Mouse IgG2B isotype control antibody (IC0041G) and Mouse Anti-Human NCAM-1/CD56 Alexa Fluor® 647-conjugated Monoclonal Antibody (FAB24086R). THP-1 human acute monocytic leukemia cell line was stained with Mouse Anti-Human LAMP-1/CD107a Alexa Fluor® 488-conjugated Monoclonal Antibody (Catalog # IC4800G, filled histogram) or isotype control antibody (IC0041G, open histogram). To facilitate intracellular staining, cells were fixed with Flow Cytometry Fixation Buffer (FC004) and permeabilized with Flow Cytometry Permeabilization/Wash Buffer I (FC005).
