Detection of Human, Mouse, and Rat DGK‑ gamma by Western Blot.
Western blot shows lysates of HeLa human cervical epithelial carcinoma cell line, MDA-MB-468 human breast cancer cell line, C2C12 mouse myoblast cell line, C6 rat glioma cell line, and SH-SY5Y human neuroblastoma cell line. PVDF Membrane was probed with 0.5 µg/mL of Mouse Anti-Human/Mouse/Rat DGK- gamma Monoclonal Antibody (Catalog # MAB6389) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF007). A specific band was detected for DGK- gamma at approximately 90 kDa (as indicated). Background: DGK-gamma
DGK-gamma (Diacylglycerol kinase gamma) is a 90 kDa, 791 amino acid (aa), type I member of the eukaryocytic diacylglycerol kinase family of enzymes. DGK-gamma possesses EF-hand, C1/Cys-rich zinc finger and catalytic domains, and is highly enriched in the retina and in cerebellar Purkinje cells. Expression elsewhere is limited to inactive isoforms missing kinase domain aa 451‑475, or zinc finger aa 333‑372. Upon calcium and PKC gamma-mediated activation, it translocates to the plasma membrane. DGK-gamma is also found in the nucleus, where it may promote cell cycle progression. Over aa 1‑126, human DGK-gamma shares 83% and 79% aa identity with mouse and rat DGK-gamma, respectively.
