Scientific Data Images for Human/Mouse/Rat STIM1 Antibody

Detection of Human, Mouse, and Rat STIM1 antibody by Western Blot.

Detection of Human, Mouse, and Rat STIM1 by Western Blot.

Western blot shows lysates of K562 human chronic myelogenous leukemia cell line, Jurkat human acute T cell leukemia cell line, CTLL-2 mouse cytotoxic T cell line, and Nb2-11 rat lymphoma cell line. PVDF membrane was probed with 1 µg/mL of Mouse Anti-Human/Mouse/Rat STIM1 Monoclonal Antibody (Catalog # MAB6976) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF007). A specific band was detected for STIM1 at approximately 86 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
Western Blot Shows Human STIM1 Antibody Specificity by Using Knockout Cell Line.

Western Blot Shows Human STIM1 Specificity by Using Knockout Cell Line.

Western blot shows lysates of HeLa human cervical epithelial carcinoma parental cell line and STIM1 knockout HeLa cell line (KO). PVDF membrane was probed with 1 µg/mL of Mouse Anti-Human/Mouse/Rat STIM1 Monoclonal Antibody (Catalog # MAB6976) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF018). A specific band was detected for STIM1 at approximately 75 kDa (as indicated) in the parental HeLa cell line, but is not detectable in knockout HeLa cell line. GAPDH (Catalog # MAB5718) is shown as a loading control. This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background: STIM1

STIM-1 (stromal interaction molecule 1), previously called GOK, is a 90 kDa type I transmembrane protein of the endoplasmic reticulum (ER). When STIM-1 senses depletion of calcium in the ER via its EF-hand domain (aa 63-98), it interacts with the plasma membrane Ca2+ release-activated Ca2+ (CRAC) channel Orai1, increasing Ca2+ influx. The human STIM-1 extracellular/lumenal domain (aa 23-213) contains an EF hand Ca2+-binding motif and a SAM (sterile a-motif) multimerization domain. A potential isoform is truncated at aa 491. Defects in STIM1 are the cause of immune dysfunction with T-cell inactivation due to calcium entry defect type 2 (IDTICED2). Within the region used as an immunogen, human STIM1 shares 95% amino acid identity with mouse and rat STIM1.
