Detection of Perforin in NK cells by Flow Cytometry.

NK cells expanded and activated (CCM032) for 14 days were harvested and stained with Mouse Anti-Human NCAM-1/CD56 APC-conjugated Monoclonal Antibody (FAB2408A) and Mouse Anti-Hunan CD3 Alexa Fluor® 405, and either (A) Mouse Anti-Human Perforin Alexa Fluor® 750-conjugated Monoclonal Antibody (Catalog # IC103853S) or (B) Mouse IgG2A Alexa Fluor® 750-conjugated Isotype Control (IC003S). Recommended Usage

Intracellular Staining by Flow Cytometry

Titration recommended for optimal concentration with starting range of 0.1-1 µg/1 million cells. Samples used for this experiment were NK cells expanded and activated (Catalog # CCM032) for 14 days were fixed and permeabilized with FlowX FoxP3 Fixation & Permeabilization Buffer Kit (Catalog # FC012) and PBMC lymphocytes fixed and permeabilized with FlowX FoxP3 Fixation & Permeabilization Buffer Kit  (Catalog # FC012).

Background: Perforin

Perforin is a pore forming, cytolytic protein that is a component of cytolytic T-cell and NK cell granules.  Perforin activates clathrin- and dynamin-dependent endocytosis, which removes perforin and granzymes from the plasm membrane to early endosomes, preserving outer membrane integrity.  Perforin facilitates granzyme uptake and avoids the proinflammatory necrotic death of a membrane-damaged cell.  Defects in the Perforin gene results in Familial hemophagocytic lymphohistiocytosis (FHL).  Perforin-based effector systems are involved not only in the lysis of abnormal cells but also in the down-regulation of cellular immune activation.
