Detection of Human PMEL by Western Blot.

Western Blot shows lysates of A549 human lung carcinoma cell line. PVDF membrane was probed with 2 µg/ml of Mouse Anti-Human PMEL Monoclonal Antibody (Catalog # MAB11470) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF018). A specific band was detected for PMEL at approximately 70 kDa (as indicated). This experiment was conducted under reducing conditions and using Western Blot Buffer Group 1.

Detection of PMEL in SK-MEL-28 cells (Positive) and K562 cells (Negative).

PMEL was detected in fixed SK-Mel-28 human malignant melanoma cell line (Positive) and absent in K562 human chronic myelogenous leukemia cell line (Negative) using Mouse Anti-Human PMEL Monoclonal Antibody (Catalog # MAB11470) at 8 µg/ml for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Mouse IgG Secondary Antibody (red; Catalog # NL007) and counterstained with DAPI (blue). Background: PMEL

Premelanasome protein (PMEL) is a nonmutated "self" antigen expressed by melanocytes, most melanoma cells, substantia nigra, and retinal pigmented epithelium.  This protein plays an essential role in the structural organization of premelanosomes. PMEL is over-expressed in all stages of melanoma progression and is negative prognostic marker for the overall survival rate of melanoma patients.  PMEL is also associated with melanin-based pigmentation, neurodegenerative disease (functional amyloid) and glaucoma (missense mutations). How PMEL amyloid is tolerated within cells is of major therapeutic interest as it is related to Alzheimer's and Parkinson's disease.
