Detection of Human RLK/TXK by Western Blot.
Western blot shows lysates of Jurkat human acute T cell leukemia cell line and human primary differentiated Th2 cells. PVDF Membrane was probed with 1 µg/mL of Human RLK/TXK Monoclonal Antibody (Catalog # MAB6520) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF007). Specific bands were detected for RLK/TXK at approximately 58 and 65 kDa (as indicated). Background: RLK/TXK
RLK (resting lymphocyte kinase), also called TXK (T cell-expressed kinase) or PTK4, is a protein tyrosine kinase of the TEK family. It is expressed in T cells and some myeloid cells, especially in Th1 cells in inflammatory diseases such as rheumatoid arthritis and Behcet’s disease. Upon TCR stimulation, it translocates to the nucleus and induces IFN-gamma production. An alternate start site creates a resident nuclear 55 kDa form in addition to the larger (~58 kDa) form that is cytoplasmic or targeted to the plasma membrane via palmitoylation of a cysteine string motif. The region used as an immunogen is common to both forms and shares 79% and 81% amino acid identity with mouse and rat RLK, respectively.
