Recommended Usage

Direct ELISA

This antibody can be used at 0.1 μg/mL with the appropriate secondary reagents to detect human VDR.

Immunohistochemistry

This antibody can be used at 20 - 40 μg/mL with the appropriate secondary reagents to detect human VDR.

Immunoprecipitation

Optimal dilutions should be determined by each laboratory.

Western Blot

This antibody can be used at 1 μg/mL under reducing conditions with the appropriate secondary reagents to detect human VDR.

This antibody is stable for greater than six months when held at -20 °C in a manual defrost freezer or at -70 °C. Upon thawing, the antibody can be stored at 2-8 °C for at least 1 month without detectable loss of activity. Avoid repeated freeze-thaw cycles.

Background: VDR/NR1I1

Vitamin D Receptor (VDR; NR1I1) is a member of the Steroid Receptor family related to the Pregnane X Receptor (PXR) and Constitutive Androstane Receptors (CARs). The natural ligand of VDR is 1, 25 di-hydroxyvitamin D3. VDR is expressed in osteoblasts, osteocytes, osteoclasts, bone, bone marrow, thymus and small intestine. VDR plays critical roles in calcium homeostasis, bone development and mineralization, as well as control of cell growth and differentiation. Retnoid X Receptors (RXRs) are the major partners for VDR since by heterodimerizing with VDR, they increase their DNA-binding affinity and select the correct spacing of direct repeat elements.
