0.2 mg/ml

Product Specifications

Immunogen

Stimulated human leukocytes

Localization

Cell surface

Specificity

Recognizes the D1 domain of an N-glycosylated glycoprotein of 120kDa with intra-chain disulfide bonds, identified as CD50 or ICAM-3. CD50 is the major ligand for LFA-1 (CD11a/CD18) and may have signaling role to increase adhesion. It is expressed on thymocytes and T lymphocytes and is resistant to treatment with phosphatidylinositol (PI) phospholipase C. This monoclonal antibody is excellent for staining of formalin/paraffin tissues.

Clonality

Monoclonal

Host

Mouse

Isotype

IgG2b Kappa

Description

200ug/ml of antibody purified from Bioreactor Concentrate by Protein A or G. Prepared in 10 mM PBS with 0.05% BSA & 0.05% azide. Also available WITHOUT BSA & azide at 1.0 mg/ml. (NBP2-34776)

Antibody with azide - store at 2 to 8C. Antibody without azide - store at -20 to -80C.

Applications for ICAM-3/CD50 Antibody (SPM505)

Application
Recommended Usage

Flow Cytometry

1-2 ug/million cells

Immunocytochemistry/ Immunofluorescence

1-2 ug/ml

Immunohistochemistry-Paraffin

1-2 ug/ml

Western Blot

1-2 ug/ml
Application Notes
Immunohistochemistry (Formalin-fixed): 1-2ug/ml for 30 minutes at RT. Staining of formalin-fixed tissues requires heating tissue sections in 10mM Tris with 1mM EDTA, pH 9.0, for 45 min at 95C followed by cooling at RT for 20 minutes.
Optimal dilution for a specific application should be determined.

Background: ICAM-3/CD50

All ICAM proteins are type I transmembrane glycoproteins, contain 2-9 immunoglobulin-like C2-type domains, and bind to the leukocyte adhesion LFA-1 protein. This protein is constitutively and abundantly expressed by all leucocytes and may be the most important ligand for LFA-1 in the initiation of the immune response. It functions not only as an adhesion molecule, but also as a potent signalling molecule. ICAM 3 is the major ligand for LFA-1 in the resting state.
