Description

This product is an IgG fraction antibody purified from monospecific antiserum by a multi-step process which includes delipidation, salt fractionation and ion exchange chromatography followed by extensive dialysis against the buffer stated above

Store this antibody at 4C prior to restoration. For extended storage aliquot contents and freeze at -20C or below. Avoid cycles of freezing and thawing. Centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4C as an undiluted liquid. Dilute only prior to immediate use.

Western Blot: IL-6 Antibody [NBP1-77894] - Shows detection of recombinant mouse IL-6 raised in E.coli. Recombinant truncated protein (0.1 ug, 21.7 kDa) was loaded on to an SDS-PAGE gel, and after separation, transferred to nitrocellulose. The membrane was blocked with 1% BSA in TBST for 30 min at RT, followed by incubation with Anti-Mouse IL-6 antibody diluted 1:1,000 in 1% BSA in TBST overnight at 4C. After washes, the blot was reacted with secondary antibody DyLight (TM) 649 conjugated Anti-Rabbit IgG (H&L) (Goat) Antibody diluted 1:20,000 in blocking buffer for 30 min. at RT.

Immunohistochemistry: IL-6 Antibody [NBP1-77894] - Shows nuclear positivity of islets of Langerhans (brown staining) and cytoplasmic staining in mouse pancreas at 10x and 20x (B & C). Staining was performed on Leica Bond system using the standard protocol. Formalin fixed/paraffin embedded tissue sections were subjected to antigen retrieval with E1 (Leica Microsystems) retrieval solution for 20 min and then incubated with rabbit anti-mouse IL-6 antibody at 1:50 dilution for 60 minutes. Biotinylated Anti-rabbit secondary antibody was used at 1:200 dilution to detect primary antibody. The reaction was developed using streptavidin-HRP conjugated compact polymer system and visualized with chromogen substrate, 3’3-diamino-benzidine substrate (DAB). The sections were then counterstained with hematoxylin to detect cell nuclei.

This purified antibody has been tested for use in Immunohistochemistry and western blotting and is suitable for ELISA. Specific conditions for reactivity should be optimized by the end user. Expect a band approximately 21.7 kDa in size corresponding to the mature 187 amino acid mouse IL-6 protein by western blotting in appropriate cell lysate or extract.

Background: IL-6

IL-6 is a multifunctional cytokine that has an important role in host defense by regulating immune and inflammatory responses. It is expressed by a variety of normal and transformed cells including T cells, B cells, monocytes/macrophages, fibroblasts and various tumor cells. IL-6 has diverse biological functions, it stimulates B-cell differentiation and antibody production, synergizes with IL-3 in megakaryocyte development and platelet production, induces expression of hepatic acute-phase proteins, and regulates bone metabolism. Recombinant human IL-6 is a 20.9 kDa protein containing 184 amino acid residues. Human IL-6 is active on murine cells but murine IL-6 is inactive on human cells.
